Distilled water is neutral. You can test it by dipping the litmus paper into it. It shows no colour change when dipped in distilled water. Therefore it is neutral.

Calamine solution is applied when an ant bites because when ant bites it injects an acid into our body. Calamine solution is basic in nature therefore it neutralizes the effect of acid that ant injects into our body.

When an ant bites it basically injects formic acid into the skin. Calamine solution contains zinc carbonate and is basic in nature. When applied on the skin, It neutralizes the affect of formic acid providing relief.
