Aim :

To observe the different stages of mitosis in onion root tip.

Apparatus :

  • Slide
  • microscope
  • cover slip
  • dilute HCl
  • spirit lamp
  • glass rod
  • needle
  • brush
  • fresh onion roots

Procedure :

➜ Collect freshly grown onion root tips cut off the last 5mm of it and place them in a test tube.

➜ Add dil. HCl and heat it on spirit lamp for 5min. to soften the root tissue.

➜ Decant the liquid carefully, wash the root tip on a watch glass containing water to remove acid.

➜ Remove the excess water with help of filter paper.

➜ Place the tip on the slide and add one drop of methylene blue.

➜ Now gently tap the root with end of the glass rod until it is completely broken.

➜ Place a cover slip on tip and warm gently for a few seconds over very low flame.

➜ Take care that it must not boil.

➜ Put a piece of filter paper over the preparation and press the cover slip with your thumb avoiding any side way movements.

➜ Observe under microscope and draw what you have observed, in your notebook.

•In this method, 2 plants of the same species but different characteristics can be cross-pollinated.

•The pollen from one plant having the characteristic of colourful flower can be used to pollinate another plant that bears big fruits.

•The resulting zygote will have the characteristics of both the pants.
