Test tube method
● Blood group systems -
- Currently various systems are being used to classify blood groups.
- Most commonly used are ABO system, Rh blood group system, MNS system, etc.
● Landsteiner's law -
- If an antigen is present on patient's red blood cells, the corresponding antibody will not be present in patient's plasma under normal conditions.
● Test tube method -
- gold standard for ABO grouping
- involves incubation of antigen-antibody for longer duration
- this test can detect weaker antigen/antibody.
Test tube method consists of 2 steps -
- cell grouping (forward grouping)
- serum grouping (reverse grouping)

"i) In medicine world, gold standard type of blood grouping is considered as the most standard and accurate method under some critical conditions.
ii) It does not contain any negative results. It clearly defines the individual blood cells and the diseases in them.
iii) Test tube method is the most suggested method in gold standard because the incubation of antibody is longer in this process and it won’t allow the antigens to dry in this process.
iv) This method also helps to identify the weak antigens in the body.
v) It is the 100% efficient and sensitive method with respect to diseases."
