1 )Fingertip has to be pierced with the help of sterile lancet and one drop of blood has to be added in each of the cavities.

2)One drop of antiserum has to be added into each cavity of the slide.

3)The blood has to be mixed with antiserum solution using mixing stick.

If there is agglutination in the form of red granule in the cavity that is because antigen reacts with the antibody.

Hence if you have AB + blood group agglutination will be seen in all chambers.

If you have O+ blood group agglutination will occur only in the Rh D cavity
