The steps of preparation/procedure are as follows-
(i)Select a plant of Balsam or Sadabahar and pluck a fresh turgid leaf.
(ii)Remove the thinnest and transparent peels of a leaf by carefully scratching with the help of a razor blade.
(iii)Pour some amount of distilled water in watch glass.
(iv)Now, put the leaf peel into the distilled water containing watch glass by a camel brush.
(v)Add a few drops of vital stain safranin with the help of a dropper into the watch glass and stain the peel.
(vi)Let the leaf peel remain in the safranin stain for at least 2 to 3 minutes for the proper staining .
(vii) A few drops (2 to 3) of glycerine are released at the center of a glass slide with the help of a dropper.
(viii)Carefully place the stained peel of the leaf on the glycerine containing glass slide.
(ix)A cover slip is placed gently on the stained material by a needle.
(x)Extra amount of glycerine should be removed from the periphery of
cover slip with the help of a blotting paper.
(xi)The glass slide along with stained specimen is placed on the stage of the compound microscope for the observations.
(xii)First of all, focus the specimen under low power(10 x) of a compound microscope to observe the stomata, guard cells and epidermal cells.
(xiii)Now, observe and identify the contents of guard cells under high power of magnification(45 x) of the compound microscope.
(xiv)Count the number of stomata,guard cells and epidermal cells.
(xv)Draw a neat and clean diagram of a stoma and label its different parts.
