Limitations of polymerase chain reaction

• PCR is much error prone as Taq polymerase has no Proof reading property.

• PCR produced DNA is small in size when the size of the cell mediated cloned DNA may reach to about 2 Mb.

• PCR is too expensive and time consuming.

• PCR cloned DNA cannot be used again and again.

• PCR does not give satisfactory result if it is not carried out with great care and in controlled condition.

Applications of PCR

• Increase in quantity of DNA sequence.

• Detection of the sight of the presence of a segment of DNA

• Disease diagnosis
