I think 2 and 4 are correct

let's take an example:-

suppose you are an IPS officer and you have been assigned to solve a mysterious case to which all were giving name as 'suicide'

you saw some red lines in accused's neck. Now, there will be two possibilities that either it was suicide or someone else has killed the accused and made the scene to seem like suicide .

you wanted to examine the cloth that was been used to hand the body around the neck

After forensic report you got a proof that there is someone's fingerprint which can be either the accused's fingerprint or the murderer's

Now you take DNA sample and match the fingerprint and voilà.

Last but not the least, you cannot put the murderer behind the bars until and unless you give proofs to court.
