Vacuole are sap filled vesicles in the protoplasm.  They are not living parts of the cell. They are used to store nutrients or waste that is to be secreted by the cell.  They are surrounded by the Tonoplast semipermeable membrane.  Tonoplast permits the exchange of solutes across it and also gases, between protoplasm and the fluid inside vacuole.
