By saying "shuffled off this mortal coil" Hamlet is trying to say- let this mortal body die.
Hamlet is very depressed due to his father’s death. The other thing tortures him that his mother marries his uncle immediately after his father’s death. The things get worse when he sees his father’s ghost who tells him that his uncle has murdered his father to cease the throne and win the queen.
The above quote is in his famous soliloquy ‘To be or not to be’. After such a catastrophe he feels everything useless and goes into depression. His wish is to get rid of his mortal body (mortal coil) and die.
