The S Phase of Interphase. The S phase of a cell cycle occurs during interphase, before mitosis or meiosis, and is responsible for the synthesis or replication of DNA. ... S phase is halted by a protein called p16 until this happens.

In the S-Phase, the most important part of cell cycle, the DNA (genome) replicates. It is then followed by division of the cell. If the s-phase fails to occur, a checkpoint before the division phase prevents the further proceedings of the cell cycle. Hence without the happening of S-phase, cell division will never complete. So , S phase is very important in the interphase of cell cycle.
