What happens if you reverse crystal violet and safranin stains in gram staining?

Both bacteria types would be stained by the safranin. When the iodine is added, safranin would be "set" in the positive. The decolorizer would wash out the safranin and then application of the crystal violet would stain the positive.

  • The crystal violet is the primary stain for gram staining procedure, it is the beginning stain for the gram positive bacteria. This imparts purple color or violet color to the bacteria.
  • A counter stain safranin is added which imparts red color to the gram negative bacteria.
  • If you reverse the staining procedure that is using safranin first, this will cause all the bacteria to remain red and crystal violet applied later on may cause the gram-negative bacteria to become violet in color and the gram -positive bacteria will remain red.
  • This will give false results for the test as crystal violet stain is used to color the gram-positive bacteria a purple-violet color.
