An oligopeptide is a short-chain peptide, i.e., a polymer of ▶amino acids (AAs) connected by amide, or more precisely peptide, linkages. The term is usually limited to peptides with less than 20–25 amino acid residues.

Oligopeptide isolation and purification refers to the process of oligopeptide enrichment, purification, and identification with gel chromatography, ion exchange chromatography, HPLC, and HPLC-MS by adopting a particular functional activity as the evaluating indicator. Gel filtration chromatography is a chromatographic method for gel isolation on the basis of different molecular masses of substances. The material used for gel filtration chromatography is a bead-like particle substance with a 3D network structure and uniform mesh diameter. When solution containing compounds with different molecular masses go through the gel column, the buffer, small compounds, can freely diffuse and permeate through the mesh while macromolecular substances are blocked outside the mesh. Substances with high molecular mass are not able to diffuse so easily through the mesh and thus are eluted earlier than substances with low molecular mass.
