The most basic characteristics of vectors are:

  • The vector needs to be a DNA molecule so that it can be cloned with the gene of interest.
  • The vector needs to have unique restriction sites. If it possesses too many restriction sites then it would be fragmented into several pieces.
  • The vector needs to have a selectable marker. The selectable markers are required to screen out transformants, non-transformants, recombinants, and non-recombinants from the colonies. Example: tet^R, amp^R, auxotrophic mutants etc.
  • The vector should have Ori site from where replication can begin.
  • The vector should be able to replicate into the host system and have a high copy number. The main motto of the cloning is to get high copy number.
  • The vector should be stable in the host system. If the vector is considered as foreign then the bacterial system may delete it. Due to this, most of the artificially derived vectors have bacterial origins.
  • Vectors such as cosmids (pJB8), YAC, and BAC are often preferred due to their ability to clone large fragments of DNA. Thus, the vector should be able to clone a large fragment of DNA.
  • If the vector has polylinkers (multiple cloning sites), then it can be utilized for directional cloning.
  • Certain vectors also possess promoters (such as pGEM3Z has SP6 and T7 promoters) which can help in getting the mRNA copy of the gene of interest which is cloned. These type of vector ease the hybridization process and identification of the gene fragments.
