A novel technique of complexing with a liquid ion exchanger, di-2-ethylhexylphosphoric acid (D2EHPA), has been used for isolation and purification of proteolytic enzymes like papain. Enzymes have been purified from their semi-purified commercial form and have also been isolated from biological sources like human placenta with the help of the cation exchanger D2EHPA at pH above the isoionic point of the enzyme concerned. Crude papain has been purified 19-fold and proteolytic enzyme has been isolated from placental extract with 70 fold purification.
