Membrane biogenesis, the synthesis of specific molecular components and their assembly into cellular membranes, is a critical, major activity in which cells engage throughout their existence. This chapter provides an overview of the membrane biogenesis. The main molecular components of cellular membranes are: (a) polar lipids, such as phospholipids, glycolipids, and cholesterol, organized into a continuous bilayer, fluid at the temperature of the immediate ambient; and (b) proteins, many of them glycoproteins—interacting in different ways with the lipid bilayer. The physical state and molecular architecture of cellular membranes is revealed by the formulation of fluid mosaic model. On account of their hydrophobic lipid bilayer, cell membranes function essentially as diffusion barriers, primarily for electrolytes and hydrophilic micro- and macromolecular solutes.
