The DNA cannot be copied in its normal form, so DNA helicase enzyme is required in order to open the strands of DNA and reveals the bases that act as a template for replicating the DNA. The area of the DNA that are opened by DNA helicase is called a replication fork.

The replication actually occurs at the replication fork, the two strands of DNA are exposed when it is opened. One strand is known as lagging strand and another is known as a leading strand. The leading strand is synthesized in 5'-3' prime, while that of the lagging strand is replicated in 3'-5' prime. DNA is always copied in 5'-3' direction.
