Staining is an auxiliary technique used in microscopy to enhance contrast in the microscopic image. Nucleous appear darker because of the acidic nature which is commonly stained by basic dye .

Staining is a spot of any thing like dishes...
Some region appear darker than other region because reaction of stain with oxygen.. Where oxygen react then their stain appear dark unless light... The nature of stain is acidic and nature of detergent is basic... And when we apply the detergent on stain it's colour will be change...
