In sickle cell anaemia there is substitution of Glutamic acid by valanine at sixth position of beta globin chain of haemoglobin molecule.
The mutant haemoglobin molecule undergoes polymerisation under low oxygen tension causing change in shape of RBC from biconcave to Sickle shaped.
