The replicon is comprised of the origin of replication (ORI) and all of its control elements. The ORI is the place where DNA replication begins, enabling a plasmid to reproduce itself as it must to survive within cells.

Origin of replication is the site from where replication starts and promoter is the region from where transcription starts.

Explanation:

  • Origin of replication is present in the DNA and this is the site from where the replication of DNA starts. Origin of replication in DNA is AT rich and 245 bp long in prokaryotes.
  • Prokaryotes contains only one origin of replication but eukaryotes contains multiple origin of replication. Promotor is the  cis acting and position dependent sequence which is responsible for starting the transcription of DNA.
  • RNA polymerase binds to promoter region and starts the transcription of genes.
