Of course, there is a major difference in the purpose.
Guru Purnima is celebrated on the 'purnima' of 'Asadh' month (according to the Hindu calendar). The 'Guru' (the spiritual guide) is honoured and gifts etc. are offered. The day is also called as Vyasa Purnima (birthday of Veda Vyasa). So the day is of religious importance.
Teacher's day on the other hand is more of an 'appeasement politics' card, the congress is so good at playing. No doubt Dr. Radhakrishnan was a great teacher, but because he was appointed as the President by the congress, it was no surprise to let 5th September be declared as a Teacher's Day, to gain the votes of teachers. That was the time when government schools were opened throughout India and congress took the opportunity to increase its vote bank.
