What is the plasmid?what is meant by ada deficiency ?how is gene therapy a solution to this problem? why is it not a permanent cure?

plasmid is an extra chromosomal circulr DNA present in some prokaryotes
it give them some unique properties like Antibiotic resistance etc

ADA defeciency means Adenosine deaminase defeciency.this enzyme is crucial for the immune system to function. the disorder is caused due to deletion of the gene for Adenosine deaminase

this is often treated through gene therapy,
lymphocytes frm the blood of patients are grown inculture medium outside the body. a functional Ada cDNA is then introduced to this lymphocytes( using retero viral vectors) , which are subsequently return to patients

how ever, as these cells are not immortal, patient require periodic infusion of such genetically engineered lymphocytes

however , if the gene isolated frm marrow cells producing ADA is introduce into cells at early embryonic stage,it could be a permenant cure
