1.- It avoids the drying of the specimen. Besides, glycerine tends to reflect light due to its refractive nature. As a result of it, image appears clearer under the microscope.
2. safranin- Because cells typically have negatively charged cell walls, the positive chromosomes in basic dyes tend to stick to the cell walls, making them positive stains.

Put the epidermis in the watch glass containing distilled water. ... Let this remain in the Safranin solution for 30 seconds, so that the peel is stained. Take the peel from the Safranin solution using the brush and place it in the watch glass containing the distilled water.
