● Activity : -

To perform mitosis in onion root tip.


● Items required : -

Two beaker on same quantity, two onion of same size (with its roots), one scissor, water.


● Procedure : -

Take two beakers. Label it as A and B. Fill half of the beaker with water. Put the onions in the beakers. Leave the apparatus for three days. Cut the root of onion place over beaker B. Again leave the apparatus for 3 days.


● Observations : -

We will observe that the onion place over beaker one has longer roots. Whereas, in beaker B the roots stoped growing.


● Process : -

Here, B onion has stoped growing beacause the meristematic tissues which helps in the growth of the plant presents in the growing parts of the plant. Such as, girth of the plant, roots and leaves. If we cut the meristematic tissue it will stop the growth.


● Conclusion : -

Therefore, we conclude that for the mitosis or division of cells in the root tip i performes by meristematic tissues. It will stop growing when the plant is fully mature and it will turn into permanent tissues.

I will follow the following procedure to observe stages of mitosis in onion root tip:

  • Collect crisply developed onion root and cut the last 5mm of it and place it in a test tube.  
  • Add dil. HC/and warmth it on heat for 5 min to diminish the root tissue.  
  • Decant the fluid cautiously; wash the root tip on a watch glass containing water.
  • Remove the abundance water with assistance of filter paper.  
  • Place the tip on the slide and include one drop of Methylene blue.  
  • Now tenderly tap the root with end of the glass bar until it is totally broken.  
  • Place a spread slip on tip and warm tenderly for a couple of moments over exceptionally low fire. Take care that it must not contain any bubbles.  
  • Put a bit of channel paper over the slide and press the spread slip with your thumb.
  • Observe under microscope and record the observation.
