If the cell in the question is the muscular cell then, the answer is D as Vesicles containing acetylcholine should reach the presynaptic membrane(neuron membrane) to be released to the synapse and go into the postsynaptic membrane(muscular membrane)

As for the gates both of them cannot open at the same time as far as I guess so.
