Here is the answer:
The number 6 glass will be filled first.

You must be thinking how?
So, when we will start putting the drink to the glass number 1, first it will go to 3 as the drink will be filled from the bottom, then it will not go to the number 7 as there is not hole and it will directly go to glass number 6

Hey, here you can see some pipes are open and close. Usually the glass that is at bottom fill first.

However, to make us think and observe details this question is made tricky. If you clearly observe the picture, the bottle itself is not opened. Hence, no glass is filled in this case.

This riddle is more on observing details, rather than thinking logically.
