In principle the following formula is correct and will apply the conditional format if D20 = "E", F20 is blank and Q20 is blank.
=AND(D20="E", ISBLANK(F20), ISBLANK(Q20))
However, when applying conditional formatting, the formula is written as if it applies to the first cell of the "Applies to" range and Excel automatically applies the formula to the remaining cells in the range.
In your formula all of the references are in relative format therefore the cells on the next row below (row 21) the formula is referencing D21, F21 and Q21.
On the next column across the formula would be referencing E20, G20 and R20.
Therefore you need to think about the absolute referencing. ie. whether just the columns need to be absolute or just the rows need to be absolute of both columns and row needs to be absolute. (Absolute is with the $ signs. Check Help for mote information on relative and absolute referencing.)
