Which technique pe used for the Separation of leaf pigment

Answer : By leaf pigment, I am assuming you mean Chlorophyll. In the lab, when we are preparing to test a leaf for the presence of Starch, we first put the leaf in a beaker, add water and heat ( perhaps using a Bunsen Burner ) until the water boils. This stops any reactions that are going on in the leaf, as the high temperature denatures any Enzymes present in the leaf. The leaf is dried and then added to some Ethanol in a beaker. The beaker is then heated with an Electric Heater ( Ethanol is highly flammable and exposing it to the naked flame of a Bunsen Burner poses a high fire risk ), until the Ethanol boils. This causes the Chlorophyll to move out of the leaf and into the Ethanol Solution, causing the leaf to decolourise from a dark green colour to a pale yellow colour.

The leaf is now ready to be tested for Starch, using Iodine Solution. We do this procedure before testing a leaf for Starch, as testing a leaf with Chlorophyll in it might cause the blue - black colour of a positive Iodine Solution Starch Test to be masked. However, since this is not required by the question, it is not a necessary component of the technique used for the removal of Chlorophyll, which has been outlined in the first paragraph.

Paper chromatography is the technique used for seperation of leaf pigment.

Mostly lreaves are green due to the presence of a pigment known as chlorophyll.

These can be seperated by using different molecules
