In the first studies on differential centrifugation of cell particles, the acid phosphatase activity of the cell was found to be contained in the mitochondrial fraction; however, if the force of centrifugation was not quite so strong the acid phosphatase activity was found to be present in the microsome fraction thus indicating that this enzyme was contained in a body of intermediate density. Subsequently it was found by De Duve that these particles also contain cathepsin (a proteolytic enzyme), acid ribonuclease, acid desoxyribonuclease, and B glucuronidase. De Duve described these particles as “lysosomes” and referred to them as “suicide bags,” since in a damaged or dead cell the enzymes would be discharged from the bodies that contain them and would autolyze the cell.
