The stomata and guard cells are seen under microscope. By using the knife the leaf cut at the centre as straight form. Next to this cut a thin, vertical slice as thin as possible of the leaf.

Then this is obtained only on above repeated steps:

  • Add few drops of water in the microscopic slide and place a “thin sliced leaf section” in water.
  • The leaf section is placed in towards to make viewers to see correct thin layer. The cover slip is added.
  • Now placing the slide under microscope with highest manifestation possible.
  • The observation is noted hereafter and hence stomata and guard cell are seen.
