Ernest Barker attended Balliol College in Oxford and Manchester Grammar School after being born in Woodley, Cheshire. From 1898 to 1905, Barker attended Merton College in Oxford. From 1909 to 1913, he attended St. John's College in Oxford.

From 1913 to 1920, he attended New College in Oxford. He attended the London School of Economics for a limited period of time.  He served as King's College London's principal from 1920 to 1927, after which he was appointed Professor of Political Science at the University of Cambridge, becoming the first occupant of the Rockefeller Foundation's chair.

He was chosen to serve on the Liberal Party Council in June 1936.

In 1944, he was knighted. The American Academy of Arts and Letters elected him as a foreign honorary member .
