They used inactivated Sendai virus to fuse the cells which were reaching separate stages in the cell cycle.
The resulting heterokaryons had two different nuclei, one from each of the parent cells but had the same cytoplasm.
They then monitored the behavioral patterns of the nuclei using microscopy and measured the M phase duration. Radiolabelling the new DNA to follow S phase was done.
