The phenomenon of UV absorbanceincreasing as DNA is denatured is known as the hyperchromic shift. The purine and pyrimidine bases in DNA strongly absorb ultraviolet light. Double-stranded DNA absorbs less strongly than denatured DNA due to the stacking interactions between the bases.
