Amazon basin is thinly populated because it's dense rain forest. Rain forests aren't a very friendly environment for human habitation, i.e. lack of fresh water, dangerous animals, lack of solid ground, lack of dry shelter, skeeters, dense trees, limited fuel and food sources, and difficult transportation.
