Why chilled ethanol is needed for dna precipitation?

we break the cell wall using cellulase or chitinase, RNA is removed by ribonuclease , proteins are removed byproteases, and we get fine threads of DNA after addition of chilled ethanol.

DNA ( Deoxy Ribo Nucleic Acid) are molecules that carry genetic matter in living things. They can be extracted using a method called DNA precipitation, in which Ethanol plays a vital role.

Benefits of using cold ethanol (about 4 degree celcius) in DNA precipitation:

  • High yield of DNA is possible.
  • Nucleic acid is kept safe by inhibiting the enzyme activity that could break it.
  • DNA molecules move from state of higher to low energy and form a precipitate. This precipitation of nucleic acid (i.e) pulling out of the DNA occurs fast and efficiency is more.
