Shylock swear about the'Jacob's staff' because of two reasons--

1)To give a biblical reference and to show Antonio that he is well aware of his rivals religion.

2)And to show Antonio that all profits earned by him(Shylock) is a blessing as long as men do not steal it.

Jacob was the son of Isaac and the grandson of Abraham

So, he was renamed "Israel" by God, Jacob is the forefather of the tribes of Israel (Jews),

He is one of whose descendants is Shylock.

Thus, Shylock's oath reminds the audience of his ethnicity
