Plants require large vacuoles because the vacuoles are used for storage of water which provides strength and rigidity to the plant shoot thereby allowing it to stay upright.
The animal cells don't require vacuoles for remaining upright since some of the cells (like bone cells also called osteoblasts) are surrounded by vitamins, minerals (like calcium) and proteins making it hard.
