It is because in a closed room there would be no ventilation . You would use up oxygen inside the room and give out carbon dioxide .As oxygen is necessary for all the processes of the body , you feel suffocated and also the carbon dioxide harms your body .

While breathing inside the closed room, we use up oxygen and give out carbon dioxide. Thus gradually the volume of oxygen decreases and the volume of carbon dioxide increases which makes us feel suffocated.
