Glycerine is added to the observation of leaf peel because normally organisms are rarely seen or it is difficult to observe the organisms.
After glycerine is added the organisms are easily spotted hence observed at ease.

Glycerin is a type of chemical which is used in observations to see the all parts like the cells, and their organelles ,etc ..
Which causes darkness in the leaf peel..
And as we know that during observation we are not able to see those parts of leaf peel. Hence that time glycerin is used.
