In simple organisms like amoeba the entire cell may be in direct contact with the environment. So their oxygen requirements can be met through diffusion. In multicellular organisms all the cells are not in direct contact with the environment. So simple diffusion cannot meet the requirements of all the cells.

In case of multicellular organisms only the cells present in the surface layer are in direct contact with the surrounding environment. The other cells and the internal organs are not in direct contact with surrounding environment. Thus , diffusion is insufficient to deliver oxygen to all parts of a multicellular organism like humans .
