Why is not possible to isolate the poles of magnet?

Because poles don't exist. To illustrate it imagine current going through a loop of wire. Tt creates a magnetic field around the wire with north and south poles but these poles are virtual, merely indicating on which side the filed lines come out and which side they go in.
