In humans, it has an important role in the absorption of vitamin B12 (cobalamin) in the intestine, and failure to produce or utilize intrinsic factor results in the condition pernicious anemia. Dietary vitamin B12 is released from ingested proteins in the stomach through the action of pepsin and acid.

Intrinsic factor is a glycoproteinsecreted by parietal (humans) or chief (rodents) cells of the gastric mucosa. In humans, it has an important role in the absorption of vitamin B12 (cobalamin) in the intestine, and failure to produce or utilize intrinsic factorresults in the condition pernicious anemia.

Intrinsic factor is secreted by parietal cells of the gastric glands in the stomach, where it binds with the vitamin. Thus bound, intrinsic factor protects vitamin B12 from digestion as it passes through the gastrointestinal tract and facilitates the vitamin's absorption in the ileum of the small intestine.
