We use glycerin for mounting the onion peel for the following reasons:

1. Glycerin which is popularly known as Glycerol helps in avoiding the dehydration of cells.

2. cells do not dry up.

3. As the refractive index of glycerin is 1.473 and it it tends to reflect ,light so the specimen kept looks clear under the microscope.

4. Glycerin is also used to expel air bubbles which can be found between slides.

We use glycerin before placing the coverslip on the slide, as it helps in not drying the safranin solution on the leaf peel. It helps in keeping the peel moisture. This is for the sample to mot get dried.
