Karyotyping

Mitoses suitable for karyotyping are obtained more easily from lymphocytecultures stimulated to grow by phytohemagglutinin. Cells cultures from amniotic fluid or biopsy of chorionic villiallow antenatal diagnoses. Skin fibroblastsor fetal blood samples from the umbilical cord of stillborns are also suitable. Direct examination of bone marrow or short-term cultures are the techniques of choice for hematological disorders. Short-term tissue cultures took advantage of technical improvements such as methotrexatesynchronization or collagenase digestion in the analysis of lymphomas and solid tumors.

Answer:

Karyotyping is the isolation, staining and visual examination of chromosomes to find chromosomal rearrangements. ... For karyotyping the chromosomes must be isolated from cells in the metaphase, which is the stage of the cell cycle in which the chromosomes assume their characteristic condensed, discrete shape.
