During any experiment if a leaf is not kept hydrated  then the epidermal cells [layer of cell that covers the leaf] on the leaf will dry up due to which you cannot observe it clearly under any compound microscope.

The students should put the leaf peel in water twice during the whole experiment because:

1. The epidemic cells that are present are visible. The epidemic cells are irregular in structure and doesn't have any inter cellular space.

2. Many small pores called stomata are scattered.

3. Pores are protected by guard cells each of them has chloroplast and nucleus.

4. The wall of the guard cell is very thick and strong. but the outer boundary is not so thick rather very thin.

5. The stomata that is present in the leaf can be opened or closed.
