Structure of protein is extremely important for the following reasons:
1. Primary structure helps us to analyse the sequence and number of amino acids present in the polypeptide chain.
2. Secondary structure helps us to understand the different non covalent forces stabilizing the structure. For example, H bonding in alpha helix and beta pleated sheet structure of protein.
3.Tertiary structure helps us to understand the functional capability of the protein. In this structure ,the protein is in a 3d shape and has active pockets/sites involving enzymatic functions.
4. Finally , if a protein has many subunits,
quaternary structure helps to understand the relationship between each of the subunits.

1. Sanger's method to determine the primary structure
2. Use of Dansyl chloride and Edmann degradation technique to understand the sequence and number of amino acids.
It is helpful especially in determining the N and C terminal amino acid.
