See, the only evidence of occurrence of photosynthesis in a plant is the production of starch.
When you wanna check whether your plant has carried out Photosynthesis during the experiment, you need to test for the presence of starch coming from photosynthesis during the experiment, and the previously formed starch should not interfere in your observation.
In order to prevent the pre-formed starch from interfering, you remove the starch (destarching) and tis' is best done by keeping the plant in dark, so that photosynthesis does not occur and the stored starch is used up.
