A simple activity to show that saliva contains enzymes is:

  • Take two test tubes marked A and B.
  • In test tube A, put boiled rice and in test tube B, put chewed rice.
  • Add water to both the test tubes and also 2-3 drops of iodine solution.
  • The rice in test tube A will change colour to bluish-black whereas in test tube B, the colour of rice will remain unchanged.  

EXPLANATION:

This is because chewed rice has been acted upon by the enzymes in saliva. The starch molecules have been converted to smaller molecules by the enzymes. In test tube A, the starch molecules are intact and react with iodine to give a bluish-black colour.
