To prepare a temporary mount of human cheek cells and study their characteristics.

Material Required-

Methylene blue stain, glycerine, a compound microscope, slide, cover slip, a clean spatula or a toothpick, a brush, a needle and a piece of blotting paper.

Theory-

-----------

Like plants, the body of all animals including humans is composed of cells.Unlike plant cells,animal cells do not have cell wall. The outermost covering of an animal cell is a cell membrane . The cytoplasm, nucleus and other cell organelles are enclosed in it. Epithelial tissue is the outermost covering of the most organs

and cavities of an animal body.

Procedure-

# Rinse your mouth with freshwater.

# With the help of a clean spatula or a toothpick , gently scrape the inner side of your cheek.

#Transfer the scrapped Material into a drop of water taken on a clear slide.

#With the help of a needle, spread the material uniformly .

#Add a drop of methylene blue stain.After about three minutes put a drop of glycerine over it.

#Place a clear cover slip over the glycerine. Remove the excess glycerine from the edges of cover slip with the help of a piece of blotting paper.

#Examine the slide under Microscope , first under low power (10X) and then under high power(45X).

#Draw diagram of cells as seen under the microscope. Observe and record the features.

#A large of flat polygonal cells arranged compactly are observed under the microscope field.

#Each cell is bounded by a thin cell membrane.

#A darkly stained distinct nucleus is observed in each cell.

#Cytoplasm is granular.

#Cell lack cell wall and large vacuoles.

Conclusion-

The cells observed under Microscope are animal cells as each cell has a cell membrane as outer boundary enclosing a distinct nucleus and dense cytoplasm.Cell wall and central prominent vacuoles are absent.

Precautions-

#Clean your mouth cavity with warm water before scraping the cheek.

#Scrape the cheek gently to avoid bleeding.

#Scraping should be properly spread so that the cells are widely separated.

#The cover slip should be kept gently to avoid entry of air bubbles.

#Always hold the slide by its edges to avoid fingerprints.

#Avoid overstaining or understaining of the material.
