The most important cities, or rather provinces under colonial rule in India were: Calcutta (today's Kolkata), Bombay (today's Mumbai), Madras (today's Chennai), Lahore (now in Pakistan), Shimla (present day, excluding Jammu), Karachi (also in Pakistan) and Delhi

Calcutta- the Company took on lease three coastal villages on the river Hooghly and developed it as springboard trading post.After shifting from Murshidabad,established their main trading center and,later on,capital of British India till 1911
Bombay- The biggest Mercantile Centre of the East and Capital of Bombay Provinces
Lahore- Biggest city of the Panjabi Provinces
Delhi- Not so important in the early days but after 1900 very important, became No:1 after WW1, was capital from 1911 onwards.
Madras- Capital of the Madras Presidency
Karachi- Chief town of Sindh/Quetta-Baluchistan
Dhaka- Important city for Jute and cotton textiles merchants and Muslims of Eastern India
Shimla - only reason, Summer Capital of India
In that order, till 1911 and then Delhi is most Important. 
There were a few important cantonments but they could be replaced easily.
The port cities are also important for British to export Indian products to our country

they played an important role in trade. row material were sent to England from these ports. similarly ,manufactured goods from England landed at these ports which were then re-distributed throughout india.
