The application of each of the following buffers are explained below -

(i) Citrate buffer - Citrate buffer is occasionally for RNA isolation because it minimized the base hydrolysis of RNA strands, and is mainly used for studying transcription.

  • This buffer makes mRNA purification invaluable for genomic research.
  • It is also used for antigen detection by breaking cross-links between antigens and any substances in its fixation medium.

(ii) HCO₃⁻ + H₂CO₃ buffer - The carbonate-bicarbonate ion buffer helps in keeping the body's pH in balance. It keeps the body's pH around 7.35 - 7.45.

  • When we eat anything that has higher acidic or alkaline. This buffer system is naturally present in the body that keeps the body pH in check.

(iii) NH₄OH + NH₄Cl​ buffer - Ammonium hydroxide and ammonium chloride buffer consist of a weak base and its conjugate base. It neutralizes the added base and acid.

  • It is used as an acid and base neutralizer.
